INTENDED USE:

The VAST BioImager system with its components will be located in DRO 221 at the University of Ottawa. The VAST BioImager system is for whole zebrafish, organ-level bright field imaging of 2-7 dpf sedated larvae. The system quickly loads, positions & rotates a fish within a thin-walled glass capillary to a user selectable orientation in the field of view of the on-board 10 μm resolution camera. Fish are loaded from a stirred, 50 ml sample container. Options are available for loading from a hand-held pipettor or from an autosampler for loading from multiwell plates. VAST software allows manual or automated control of all the steps needed to capture and store images of the fish in your sample. The VAST system can be placed on upright microscopes for whole fish fluorescence imaging and can be integrated with some microscopes for automated, high-content image capture. The (Large Particle) LP Sampler is an automated sample introduction system designed specifically for gentle handling of large fragile objects such as zebrafish larvae. It is capable of aspirating samples from the wells of multiwell plates and delivering them intact to Union Biometrica’s VAST BioImager system.

The system consists of one VAST BioImager™ SYSTEM and its components:

1. VAST BioImager™ SYSTEM with MICROSCOPE INTEGRATION 700-5000-000: The VAST BioImager system is for whole zebrafish, organ-level bright field imaging of2-7 dpf sedated larvae. The system quickly loads, positions & rotates a fish to a user selectable orientation in the field of view of the on-board 10 µm resolution camera. Fish are loaded from a stirred, 50 ml sample container. Options are available for loading from a hand-held pipettor or from an autosampler for loading from multiwell plates. VAST software allows manual or automated control of all the steps needed to capture and store images of the fish in your sample. The VAST system can be placed on upright microscopes for whole fish fluorescence imaging and can be integrated with some microscopes for automated, high-content image capture.LP Sampler™ 620-5000-000 and Probe 620-5002-002: The (Large Particle) LP Sampler and probe is an automated sample introduction system designed specifically for gentle handling of large fragile objects including large cells/clusters and model organisms. It is capable of aspirating samples from the wells of multiwell plates and delivering them intact to Union Biometrica’s VAST BioImager system. Software for seamlessly integrating the LP Sampler with these other systems is included. Sample object size can range from 1 to 1500 microns in diameter. This high-throughput device is capable of processing multiple samples simultaneously for continuous sample introduction.

2. Dispenser Kit for VAST 620-5024-000: LP Sampler Dispenser Kit for VAST turns the LP Sampler into an instrument capable of gently aspirating and dispensing 2-7 dpf zebrafish larvae. The LP Sampler is capable of gently aspirating larvae from 24, 48 and 96-well plates and delivering them intact to the VAST BioImagerTM system for automated imaging. After imaging is complete, this dispensing feature allows each larva to be deposited into the corresponding well of a daughter plate. This dispensing feature can also work with the VAST sample cup or the VAST Pipettor for bulk filling of multiwell plates with zebrafish that have been imaged on the VAST BioImager.

3. VASTomography Software 700-5023-000: VASTomography software for optical projection tomographic reconstructions. This software works with the VAST BioImager to create three-dimensional density models of zebrafish larva. These 3D models can be used for visualizing and quantifying information such as relative position, shape, and volume of features.

4. VAST Pipettor 700-5016-000: Hand-held, Flow-through, Pipettor for loading zebrafish to the VAST at the click of a button.

• The VAST is a capillary system where larvae can be loaded and manipulated for imaging.
•  Zebrafish larvae can be loaded into the system using the 50 mL stirred conical tube sample cup or from wells of a multi-well plate.
• The VAST loads larva one at a time into the capillary and positions it laterally in the field of view of the on-board camera.
• The system rotates each larva 360 degrees to determine the orientation of the fish. The operator can then select the desired orientation: lateral, dorsal or any other custom orientation for image capture. All the remaining larvae will be rotated automatically to the same orientation using pattern recognition algorithms.
• Once the image is captured, the software can direct the VAST unit to eject the larva to waste or dispense it to a chosen receptacle for further procedures.
• VASTomography™ Software
  • Ability to generate 3D optical projection tomography (OPT) in minutes.
  • Data sets of volume, shape, and position for selected region of interest.
• VAST screening of an entire 96 well plate, with 1 larva per well, takes on average, 1 min per well. Performed manually, assays of similar complexity require about 10 min per zebrafish, and the assays are error-prone.
