Statement of Requirements

This Request for Proposal (“RFP”) is an invitation to prospective Proponents to submit Proposals for the provision of an Automated Liquid Handler to perform large scale genome modulation screens to identify critical genetic pathways for the pathogenesis of Parkinson’s disease and ALS. The University recently acquired a high-content imager (“HCI”) and this item will bring exciting complementarity to this set-up. Together, the HCI and automated liquid handler will comprise an integral portion of the functional genomic screening pipeline for the University’s Faculty of Medicine researchers.

General Description

The requested Automated Liquid handling platform must be compact with 8-channels, and offer speed, flexibility, ease of use and superior pipetting performance for high-throughput screening experiments. The addition of the requested liquid handler to the existing molecular screening equipment at the Faculty of Medicine will represent the final touch to assemble a complete technical pipeline allowing automated, highly reliable and reproducible high-throughput screening endeavors. Specifically, the requested liquid handling system must allow precise and consistent dispensing of culture media, cell suspensions, viral particles, growth factors, and small molecules in miniaturized cellular assays. Equipped with an 8-channel dispensing head and a precision range of 1 to 1000 ml, this automated workstation must be highly versatile so it can execute several key steps of a high-throughput experimental pipeline. For instance, it must enable the preparation of a large quantity of tissue culture experimental plates demanding specific growth/adhesion matrix coating, precise cell seeding and viral transduction, as well as time-sensitive media feeding. It must subsequently perform standardized molecular treatments, washing steps, cell fixation and any other steps of a staining protocol in a time-efficient manner, prior to high-content imaging.

The Appendix A specifications contained in this RFP are the mandatory minimum requirements for the Automated Liquid Handler.

Minimum Mandatory Requirements

1. Versatility and Accuracy:
1.1 Must have a minimum of 8 sample probes.
1.2 Must have automated and adjustable spacing function between sample probes.
1.3 Must enable liquid dispensing in both 96- and 384-well plate format.
1.4 Must include troughs for washing tips.
1.5 Must include pump and liquid enclosure to eliminate carry-over.
1.6 Must include all necessary pipetting adaptors.
1.7 Must include a tip rack holder for different tip sizes.
2. Precision
2.1 Must include liquid level sensors.
2.2 Must be able to pipette as little as 1 µl and as much as 1000 µl.
3. Sterility
3.1 Must include an enclosure around the entire liquid handler (HEPA/CL2 certified), allowing for sterile procedures.
4. Other Requirements
4.1 Installation and training must be included.
4.2 Must include four years warranty on parts and labor (including standard manufacturer warranty).
4.3 Must have an automation pipeline.
4.4 Must include software, as well as computer and monitor (unless equipment has an integrated touchscreen).
4.5 The packaged shipment(s) must be able to pass through the lab doorway which measures 36-1/2″ width x 84” high.
