Description

The University requires spinning disk confocal microscope based on Nipkow disk equipped with micro lenses for fast imaging of intracellular dynamics in living cells in the brain sections.

The system must contain an upright microscope and will be used for

For imaging calcium signals propagation in the thin processes of developing cells
For imaging fast intracellular dynamics of organelles in distinct types of cells in the living brain sections

The confocal microscope based on Nipkow disk equipped with micro lenses must have clearly separated light paths for excitation and emission, and even illumination. The spinning disk should allow for at least 10,000 rpm and 50 um pinhole size. The requested system is composed of an upright microscope equipped with a microscope controller, a motorized six-position fluorescent cube filter, and an epifluorescent module with appropriate filters and dichroics for imaging DAPI, GFP, TRITC and Cy5. The system should be equipped with four laser lines at 405, 488, 561 and 640 nm and a high sensitivity camera with 95% QE a sensor size of at least 18.8mm, 6.5x6.5um, 2048x2048 array size and low read noise. The camera should allow for 95fps at full array. A high numerical aperture (NA) 60x water-dipping objective (NA, 1.0) with apochromatic correction or better with corrections for Near Infra-Red observations with a 2.8-mm working distance (WD) and a 10x objective (NA, 0.45, WD 4.0 mm) are also needed. The system should be also equipped with a piezo z stage and a motorized xy stage with encoder as well as a workstation and software package to control the entire system and provide 2D/3D deconvolution options, object tracking, and 3D measurements.
The system will be used for tracking fast intracellular processes in different cell types in the brain sections
